چکیده

of an adult wild-type mouse that received a homotopic graft of subventricular zone cells from a GFP /TrkB / mouse and was killed 15 d later. Graft-derived neurons (green) migrate to the olfactory bulb, disperse radially into several olfactory bulb layers (granular layer shown in picture; red, DAPI nuclear dye), and extend dendrites even in the absence of the neurotrophin receptor TrkB. Slit2 regulates the dispersal of oligodendrocyte precursor cells via Fyn/RhoA signaling.

Oligodendrocyte precursor cells (OPCs) are a unique type of glia that are responsible for the myelination of the central nervous system. OPC migration is important for myelin formation during central nervous system development and repair. However, the precise extracellular and intracellular mechanisms that regulate OPC migration remain elusive. Slits were reported to regulate neurodevelopmental...

Functional N-methyl-D-aspartate receptors in O-2A glial precursor cells: a critical role in regulating polysialic acid-neural cell adhesion molecule expression and cell migration

The capacity for long-distance migration of the oligodendrocyte precursor cell, oligodendrocyte-type 2 astrocyte (O-2A), is essential for myelin formation. To study the molecular mechanisms that control this process, we used an in vitro migration assay that uses neurohypophysial explants. We provide evidence that O-2A cells in these preparations express functional N-methyl-D-aspartate (NMDA) re...

The NG2 proteoglycan: past insights and future prospects.

The NG2 chondroitin sulfate proteoglycan is a valuable marker for several types of incompletely-differentiated precursor cells, including oligodendrocyte progenitors in the central nervous system, developing mesenchymal cells in cartilage, muscle, and bone, and pericytes/smooth muscle cells in developing vasculature. In addition to extending our knowledge about the developmental roles of these ...

The Chemokine Receptor CXCR2 Controls Positioning of Oligodendrocyte Precursors in Developing Spinal Cord by Arresting Their Migration

Spinal cord oligodendrocytes originate in the ventricular zone and subsequently migrate to white matter, stop, proliferate, and differentiate. Here we demonstrate a role for the chemokine CXCL1 and its receptor CXCR2 in patterning the developing spinal cord. Signaling through CXCR2, CXCL1 inhibited oligodendrocyte precursor migration. The migrational arrest was rapid, reversible, concentration ...

Calcium signaling in chemorepellant Slit2-dependent regulation of neuronal migration.

Migration of neuronal precursor cells in the developing brain is guided by extracellular cues, but intracellular signaling processes underlying the guidance of neuronal migration are largely unknown. By examining the migration of cerebellar granule neurons along the surface of cocultured astroglial cells, we found that an extracellular gradient of Slit2, a chemorepellant for neuronal migration ...
